Pan Anti-beta Amyloid scFVs, Anti-beta Amyloid scFVs Specific for Abeta-40 and Abeta-42 and Methods for Their Delivery to Treat or Prevent Alzheimer's Disease

Description

Accumulation of Abeta in the brain of patients who suffer from Alzheimer’s disease (AD) triggers a complex series of events that eventually result in neuronal dysfunction and death. Selective targeting of Abeta using high affinity binding agents holds great promise as a potential way to treat or prevent AD. In order to avoid complications associated with immune effector functions of antibodies we have generated novel recombinant single chain autobodies that selectively bind either Abeta40 or Abeta 42. Delivery of these to the brain of Abeta depositing mice significantly retards amyloid deposition. Nucleic acid and protein sequences of these scFVs are disclosed as well as methods for treatment of AD with these anti-abeta scFV are described.
