Description
We have conducted microarray chip analysis of estrogen-regulated genes in the U2OS0ERa, -ERb, and -ERa/b osteoblast cell lines to determine the relative contribution to endogenous gene expression. Numerous genes were identified that can potentially serve as markers to investigate the effects of both known and novel estrogen receptor ligands in osteoblats. Some of the ERa-specific genes include: von Willebrand Factor, mucin 1 and nuc2 homolog (inolved in cell death). Some of the ERb-specific genes include: the nuclear protein SA-1, inhibitor of apoptosis homolog C (IAP-C), angiotensinogen and deleted in colorectal cancer (DCC). In addition, cystatin S, cystatin D and interleukin-8 receptor beta were among those genes regulated by both receptor isoforms. We have found that the U2OS-ERa/b line, containing both receptor isoforms, inhibits the estrogen-induction of angiotensinogen, IAP-C and SA-1 genes.
