Description
We have replace the mouse islet amyloid polypeptide (IAPP, amylin) gene with the human IAPP gene. Human IAPP is readily converted to amyloid fibrils. Amyloids have been associated with a large number of human diseases that are associated with cell death. Mouse IAPP is non-amyloidogenic. By replacing the mouse IAPP with the human IAPP gene we have created a physiologic model of hman IAPP expression in the mouse, providing an idealized model system with which to study the influence of amyloid formation in the pathogenesis of type 2 diabetes. This model may be very useful for screening drugs that may be used to prevent and/or treat type 2 diabetes. A more detailed invention Description is attached to this disclosure.
